Abstract

Chronic wounds significantly reduce a patient's quality of life. Wound dressing is an effective method of treating wounds and promoting healing. Alginate hydrocolloids are biocompatible and absorb exudate to form a gel, which creates a moist wound environment conducive to cell proliferation. The incorporation of herbal extracts into alginate hydrogels may have the potential to accelerate the healing process. Calendula officinalis L. contains flavonoids and triterpenes, which have been shown to have anti-inflammatory properties as well as cell proliferation, accelerating wound healing. The goal of this study was to create an alginate hydrogel containing marigold extract, characterize its physicochemical properties, and assess its effect on fibroblast viability in vitro. Dried marigold flowers were used to make glycolic extracts. Glycolic extract UV-visible spectrophotometry and HPLC-DAD analysis. The presence of phenolic compounds such as flavonoids and phenolic acids was revealed by UV-visible spectrophotometry and HPLC-DAD analysis of glycolic extract. Propylene glycol:water extracting solutions ranging from 10 to 30% (v/v) in water provided the best extracting solution for the preparation of glycolic extracts. Stability tests, thermogravimetric (TG) analysis, and rheology were performed on the alginate-calendula hydrogel. The physicochemical tests revealed that the formulation is stable, has a pH compatible with the skin, and samples stored at 4 °C during storage stability testing did not exhibit significant changes in pH, viscosity, or spreadability. The rheological tests show that viscosity decreases with increasing temperature, with the viscous character (G") dominating at low frequencies. In murine fibroblasts 3T3 cells, cell viability was determined using methyl tetrazolium and sulforhodamine B assays. According to the findings, the alginate-calendula hydrogel formulation was not cytotoxic. The SRB assay revealed an increase in cell viability at dilutions beginning at 1:80.
