dc.description.abstractPhotodynamic Therapy (PDT) could be an alternative for the treatment of potentially malignant oral disease or for early-stage squamous cell carcinoma. Less costly photosensitizers could be developed and tested, including methylene blue. The aim of this research was to investigate in vitro the effect of photodynamic therapy using methylene blue as a gel in a cell line derived from oral squamous cell carcinoma (SCC- 4). The methylene blue gel was developed at a predetermined concentration and pH of 6. After preparation of the base gel, incorporation and cytotoxicity tests of the gel were performed with methylene blue at concentrations ranging from 25μm to 400μm. Cell viability was determined by experiments with dimethyltiazoldiphenyltetrazolium (MTT) to verify the efficacy of the gel. Based on the data obtained, we conclude that PDT with AM gel causes a decrease in cell viability of SCC-4 cells after 30 minutes of exposure, but without cytotoxicity.pt_BR
dc.description.resumoA Terapia Fotodinâmica (PDT) pode ser uma alternativa para o tratamento das desordens orais potencialmente malignas ou nos casos de carcinoma espinocelular em estágios iniciais. Os fotossensibilizadores com menor custo poderiam ser desenvolvidos e testados; dentre eles, o Azul de Metileno. O objetivo da pesquisa foi avaliar in vitro a ação da Terapia Fotodinâmica utilizando o Azul de Metileno na forma de gel, em linhagem de célula derivada de carcinoma espinocelular de boca (SCC-4). O gel de Azul de Metileno foi desenvolvido em concentração preestabelecida e com pH 6. Após o preparo do gel base foram realizados testes de incorporação e citotoxicidade do gel com Azul de Metileno em concentrações que variam de 25μm a 400μm. A viabilidade celular foi determinada através de experimentos com dimetiltiazoldifeniltetrazolium (MTT), buscando verificar a eficácia do gel. Com base nos dados obtidos, conclui-se que a PDT com gel de AM induz uma diminuição na viabilidade celular de células SCC-4 passados 30 minutos de exposição, porém sem apresentar citotoxicidade.pt_BR
