Abstract

Photodynamic Therapy (PDT) could be an alternative for the treatment of potentially malignant oral disease or for early-stage squamous cell carcinoma. Less costly photosensitizers could be developed and tested, including methylene blue. The aim of this research was to investigate in vitro the effect of photodynamic therapy using methylene blue as a gel in a cell line derived from oral squamous cell carcinoma (SCC- 4). The methylene blue gel was developed at a predetermined concentration and pH of 6. After preparation of the base gel, incorporation and cytotoxicity tests of the gel were performed with methylene blue at concentrations ranging from 25μm to 400μm. Cell viability was determined by experiments with dimethyltiazoldiphenyltetrazolium (MTT) to verify the efficacy of the gel. Based on the data obtained, we conclude that PDT with AM gel causes a decrease in cell viability of SCC-4 cells after 30 minutes of exposure, but without cytotoxicity.
