dc.description.abstractAcinetobacter baumannii is a Gram-negative coccobacillus, opportunistic, and emerging as a nosocomial pathogen. Its success as an infectious agent is attributed to several virulence factors, particularly its ability to form biofilms, release extracellular proteases, and develop resistance to antimicrobials. This study aimed to investigate the secretion of proteases by different strains of A. baumannii, biofilm formation, and antibiotic resistance profiles, seeking to understand the influence of these factors on the pathogenicity of this bacterium. Twenty strains from the University Hospital of Ponta Grossa were analyzed and confirmed as A. baumannii through glucose and lactose oxidation tests and growth at 42°C. Qualitative tests of proteolytic degradation revealed that, of the 20 strains analyzed, 7 (35%) showed a degradation halo for both skim milk and milk agar media, while 13 (65%) exhibited this characteristic in only one of the media. The results of the qualitative caseinolysis tests in liquid medium partially confirmed these findings, as only 20% of the samples showed minimal casein degradation. Similarly, zymography revealed the presence of enzymatic activity bands around 25 kDa and 10 kDa. The samples ABHU1 and ABHU4 showed proteolytic activity in all applied tests. Furthermore, they demonstrated satisfactory results for the biofilm formation test, being classified as strong biofilm producers. All samples were resistant to the tested antimicrobials, except for colistin. Despite the large number of studies involving the association between A. baumannii with hospital outbreaks, severe infections, and antimicrobial resistance, factors that determine the virulence and pathogenicity of this microorganism need to be studied in depth. It becomes evident that the ability to form biofilms, in addition to other characteristics of this microorganism, can be considered an important virulence factor common to a large number of A. baumannii isolates. Detailed investigations still need to be conducted to provide new data on the proteases released by different strains of A. baumannii and their role in pathogenesis.pt_BR

Seu sucesso como agente infeccioso é atribuído a diversos fatores de virulência, destacando-se sua capacidade de formar biofilmes, liberar proteases extracelulares e desenvolver resistência a antimicrobianos. Este estudo teve como objetivo investigar a secreção de proteases por diferentes cepas de A. baumannii, a formação de biofilme e o perfil de resistência a antibióticos, buscando entender a influência desses fatores na patogenicidade dessa bactéria. Foram analisadas vinte cepas provenientes do Hospital Universitário de Ponta Grossa, confirmadas como A. baumannii por meio de testes de oxidação de glicose e lactose e crescimento a 42°C. Os testes qualitativos de degradação proteolítica revelaram que, das 20 cepas analisadas, 7 (35%) apresentaram halo de degradação para ambos os meios de skim milk e ágar leite, enquanto 13 (65%) demonstraram tal característica em apenas um dos meios. Os resultados dos testes qualitativos de caseinólise em meio líquido parcialmente confirmaram esses achados, visto que apenas 20% das amostras mostraram degradação mínima da caseína. Da mesma forma, a zimografia revelou a presença de bandas de atividade enzimática em torno de 25 kDa e 10 kDa. As amostras ABHU1 e ABHU4 apresentaram atividade proteolítica em todos os testes aplicados. Além disso, demonstraram resultados satisfatórios para o teste de formação de biofilme, sendo classificadas como fortes produtores de biofilme. Todas as amostras foram resistentes aos antimicrobianos testados, com exceção à colistina. Apesar do grande número de estudo envolvendo a associação entre A. baumannii com os surtos hospitalares, infecções graves e resistência aos antimicrobianos, fatores que determinam a virulência e patogenicidade deste micro-organismo precisam ser estudados de modo aprofundado. Torna-se evidente que a capacidade de formação de biofilme, além de outras características desse micro-organismo, pode ser considerada como importante fator de virulência comum a um grande número de isolados A. baumannii. Investigações detalhadas ainda precisam ser realizadas para fornecerem novos dados sobre as proteases liberadas por diferentes cepas de A. baumannii e seu papel na patogênese.pt_BR
