dc.description.abstractEucalyptus stands out as the most cultivated globally within the Myrtaceae family, with notable morphological similarity. It presents growing economic importance due to the essential oils (EOs) produced, which are attributed to diverse biological activities. This study aimed to investigate the petiolar morphoanatomy, phytochemistry and biological activities of essential oils from leaves of six Eucalyptus species, namely: E. badjensis, E. benthamii, E. dunnii, E. grandis, E. globulus and E. saligna, collected seasonally (spring, summer, autumn and winter). Cultivation conditions and plant development stage were standardized for all species. The morphoanatomy was analyzed using standard microscopy methods. The EOs from the leaves were extracted by hydrodistillation. The chemical profile was determined by GC-EM and CCDAE and the biological activities were tested for insecticidal (topical and fumigation) and anti- inflammatory action (carrageenan-induced paw edema, mechanical hyperalgesia, sensitivity to cold and pleurisy). The morphoanatomical study of the petioles highlighted the following differences: size, cross-sectional shape, size of cavities, arrangement, and number of vascular bundles, in addition to crystalline morphotypes. The essential oils of the six species showed significant variations in the content, chemical constitution and concentration of volatile compounds depending on seasonality. Yields varied between 0.7-2.43%, being the highest for E. badjensis in summer and the lowest for E. grandis in winter. The major constituents were in E. badjensis (1,8-cineole), E. benthamii (α-pinene and globulol), E. dunnii (α-pinene and 1,8-cineole), E. globulus (1,8- cineole), E. grandis (α-pinene and o-cymene) and E. saligna (o-cymene and cryptone). High-performance thin-layer chromatography has emerged as a rapid technique for monitoring chemical composition. GC-MS and CCDAE analyzes showed that different Eucalyptus had unique chemical compositions. This finding was demonstrated in principal components cluster analysis. Insecticide analysis revealed that E. benthamii EO is the most potent against Cimex lectularius. The six EOs demonstrated antioxidant activity in the ABTS model. EOs from E. benthamii, E. dunnii and E. grandis showed significant anti-inflammatory activities in tested experimental models. These findings highlight the importance of studying Eucalyptus essential oils, not only for their chemical diversity and therapeutic potential but also for their seasonal variability and specificity between species as illustrated in the anatomical study.pt_BR

Apresenta crescente importância econômica pelos óleos essenciais (OEs) produzidos, aos quais são atribuídas atividades biológicas diversas. Este estudo teve como objetivo investigar a morfoanatomia peciolar, a fitoquímica e as atividades biológicas dos óleos essenciais de folhas de seis espécies de Eucalyptus, a saber: E. badjensis, E. benthamii, E. dunnii, E. grandis, E. globulus e E. saligna, coletadas sazonalmente (primavera, verão, outono e inverno). As condições de cultivo e estágio de desenvolvimento das plantas foram padronizadas para todas as espécies. A morfoanatomia foi analisada por métodos usuais de microscopia. Os OEs de folhas foram extraídos por hidrodestilação. O perfil químico foi determinado por CG-EM e CCDAE e as atividades biológicas foram testadas quanto ao efeito antioxidante, ação inseticida (tópico e fumigação) e anti-inflamatória (edema de pata induzido por carragenina, hiperalgesia mecânica, sensibilidade ao frio e pleurisia). O estudo morfoanatômico dos pecíolos evidenciou as seguintes diferenças: tamanho, formato em secção transversal, tamanho de cavidades, disposição e o número de feixes vasculares, além dos morfotipos cristalinos. Os OEs das seis espécies apresentaram variações significativas no teor, na constituição química e na concentração dos compostos voláteis em função da sazonalidade. Os rendimentos variaram entre 0,7-2,43% sendo o maior para E. badjensis no verão e o menor para E. grandis no inverno. Os constituintes majoritários foram em E. badjensis (1,8-cineol), E. benthamii (α-pineno e globulol), E. dunnii (α-pineno e 1,8-cineol), E. globulus (1,8-cineol), E. grandis (α- pineno e o-cimeno) e E. saligna (o-cimeno e criptona). A cromatografia em camada delgada de alta eficiência se apresentou como uma técnica rápida para monitorar a composição química. As análises por CG- MS e CCDAE mostraram que diferentes Eucalyptus apresentaram composições químicas únicas. Esse achado foi demonstrado na análise de cluster de componentes principais. A análise de inseticida revelou que o OE de E. benthamii é o mais potente contra Cimex lectularius. Os seis OEs demonstraram atividade antioxidante no modelo ABTS. Os OEs de E. benthamii, E. dunnii e E. grandis mostraram atividades anti-inflamatórias significativas em modelos experimentais testados. Esses achados destacam a importância do estudo dos OEs de Eucalyptus, não apenas por sua diversidade química e potencial terapêutico, mas também por sua variabilidade sazonal e especificidade entre as espécies como é ilustrado no estudo anatômico.pt_BR
