dc.description.abstractThe expansion of agricultural practices has caused degradation in several terrestrial ecosystems, due to the intensification of land use and conversion of native ecosystems into agroecosystems. These practices impact the structuring of soil microbial communities and, consequently, the ecosystem services provided by the soil microbiota. The Campos Gerais in Paraná, composed of extensive areas of native grassland vegetation, can be considered fragile due to exposure to abiotic factors, exclusion from public conservation policies, and intense pressure from agriculture and silviculture. Ecological restoration practices are an alternative for recovering native grassland areas degraded by anthropic actions. However, the impact that land use changes have on the microbial communities of these ecosystems is still poorly understood. For this reason, high throughput sequencing and comparative metagenomics were used to verify this composition of the taxonomic and functional microbial communities in soils from native fields with different use histories: (1) agricultural area, (2) restoration area passive ecological since 2017 and (3) native area in a preservation unit. The results show that the bacterial community of the agricultural area, followed by the restoration area, present the highest Shannon diversity index values that were significantly different from the preservation area. The assessment of the taxonomic composition of the areas revealed that phyla considered copiotrophic, such as Pseudomonadota, were more abundant in the environment in the preservation area, whereas oligotrophic phyla like Acidobacteria were more abundant in the agricultural area, in contrast, the abundance of these phyla in the restoration area decreased. The Archaea community did not show differences in Shannon index and Pielou equitability and, therefore, were more resilient to land use changes. The fungal diversity was higher in the preservation area and was significantly distinct from the agricultural area, while the restoration area was more similar to the preservation area. The beta diversity analysis calculated by the Bray-Curtis dissimilarity indicates that passive ecological restoration for five years has caused changes in the soil microbial community. Furthermore, the agricultural and restoration areas showed a greater amount of differentially abundant functional genes when compared to the preservation area, which may be responses to greater disturbances in these environments. In a principal component analysis (PCA) where PC1 was responsible for 32,6% of the variation, it was evidenced that the functional diversity found in the agricultural area is farther from the preservation area, and the restoration area is in an intermediate position, showing that the functional composition was also affected by the history of land use. These results indicate that the microbial community of anthropized soils of native fields already shows recovery of its structure in just five years of passive restoration, highlighting the importance of restoration and conservation practices of the ecosystems of Campos Gerais in Paraná.pt_BR

Práticas como estas impactam a estruturação de comunidades microbianas do solo e, consequentemente, nos serviços ecossistêmicos prestados pela microbiota do solo. Os Campos Gerais no Paraná, composto por extensas áreas de vegetação de campos nativos, podem ser considerados frágeis pela exposição a fatores abióticos, exclusão de políticas públicas de conservação e forte pressão da agricultura e silvicultura. As práticas de restauração ecológica são alternativas para recuperação de áreas de Campos Nativos degradados por ações antrópicas. No entanto, o impacto que as mudanças no uso de terra têm sobre as comunidades microbianas desses ecossistemas ainda é pouco compreendido. Com este intuito, o sequenciamento de alto rendimento e a metagenômica comparativa foram utilizados para verificar a composição da comunidade microbiana de caráter taxonômico e funcional em solos de Campos Nativos com diferentes históricos de usos: (1) área agrícola, (2) área de restauração ecológica passiva desde 2017 e (3) área nativa em unidade de preservação. Os resultados mostram que comunidade bacteriana da área agrícola, seguido da área de restauração, apresentaram os maiores valores quanto ao índice de diversidade de Shannon e foram significativamente diferentes da área de preservação. A avaliação da composição taxonômica das áreas revelou que filos considerados copiotróficos como Pseudomonadota foram mais abundantes no ambiente em área de preservação, enquanto filos oligotróficos, como Acidobacteria foram mais abundantes em solo agrícola, ao passo de que a abundância destes filos em área de restauração diminuiu. A comunidade de Archaea não apresentou diferenças quanto aos índices de Shannon e a equitabilidade de Pielou e, portanto, mostraram-se mais resilientes quanto as mudanças de uso de solo. A diversidade fúngica foi maior em área de preservação e foi significativamente distinta da área agrícola, enquanto a área de restauração foi mais semelhante a área de preservação. A análise da diversidade beta calculada pela dissimilaridade de Bray-Curtis indica que a restauração ecológica passiva por um período de cinco anos provocou alterações na comunidade microbiana do solo. Além disso, as áreas agrícola e de restauração apresentaram maior quantidade de genes funcionais diferencialmente abundantes quando comparadas com a de preservação, que pode ser respostas à maiores perturbações nestes ambientes. Em uma análise de componentes principais (PCA) onde PC1 foi responsável por 32,6% da variação foi evidenciado que a diversidade funcional encontrada em área agrícola está mais distante da área de preservação, e a área de restauração encontra-se em posição intermediária, mostrando que a composição funcional também foi afetada pelo histórico de uso do solo. Estes resultados indicam que a comunidade microbiana de solos antropizados de Campos Nativos já apresentam recuperação de sua estrutura em apenas cinco anos de restauração passiva, salientando a importância de práticas de restauração e conservação dos ecossistemas dos Campos Gerais no Paraná.pt_BR
