dc.description.abstractThe treatment of chronic wounds is a highly dynamic and variable process. There are several types of curatives for the treatment of chronic. Alginate is a biopolymer extracted from brown algae that is widely used in the manufacture of wound dressings. Alginate hydrogels absorb water and keep the wound moist, providing a suitable environment for cell growth and proliferation. The healing effect of alginate can be enhanced by natural compounds. However, there are no alginate dressings containing medicinal plant extracts with a healing effect. Aloe barbadensis Mill., popularly known as aloe vera, has anti-inflammatory and healing actions. The aim of the present study was to obtain and characterize the physicochemical properties of alginate hydrogels containing aloe vera gel and evaluate the in vitro healing effect. The formulations were submitted to evaluation of pH, viscosity, relative density, spreadability, organoleptic characteristics and accelerated stability test. An alginate hydrogel formulation containing aloe vera gel with the best profile for topical use was selected for in vitro assays: cell viability by MTT and SRB methods and cell migration by Scratch assay, using fibroblasts cells. The physical-chemical tests showed that the formulations are stable and have a pH compatible with the skin. The spreadability and viscosity values varied according to the polymeric composition of the formulations. In the in vitro cell viability assay, it was observed that the cell morphology was not altered, as well as viability and multiplication capacity of the cells. In the in vitro cell migration assay, when compared to the control, the formulation with aloe gel accelerated the cell migration process. The results confirmed the possibility of incorporating aloe vera gel in alginate hydrogels, obtaining stable formulations suitable fortopical use with potential as wound healing materials.pt_BR
dc.description.resumoO tratamento de feridas crônicas é um processo altamente dinâmico e variável. Há diversos tipos de curativos para o tratamento de lesões crônicas. O alginato é um biopolímero extraído de algas marrons e amplamente utilizado na confecção de curativos. Os hidrogéis de alginato absorvem água e mantêm a ferida úmida, propiciando um ambiente adequado para crescimento e proliferação celular. O efeito cicatrizante do alginato pode ser potencializado por compostos de origem natural. Entretanto, não há curativos de alginato contendo extratos de plantas medicinais com efeito cicatrizante. Aloe barbadensis Mill., popularmente conhecida como babosa, possui ações anti-inflamatória e cicatrizante. O objetivo do presente estudo foi obter e caracterizar as propriedades físico-químicas de hidrogéis de alginato contendo gel de babosa e avaliar o efeito cicatrizante in vitro. As formulações foram submetidas a avaliação do pH, viscosidade, densidade relativa, espalhabilidade, características organolépticas e ensaio de estabilidade acelerada. Uma formulação de hidrogel de alginato contendo gel de babosa com melhor perfil para o uso tópico foi selecionada para ensaios in vitro: viabilidade celular, pelos métodos de MTT e SRB; e migração celular, pelo método Scratch, utilizando uma linhagem de fibroblastos. Os ensaios físico-químicos evidenciaram que as formulações são estáveis e possuem pH compatível com a pele. Os valores de espalhabilidade e viscosidade variaram de acordo com a composição polimérica das formulações. No ensaio in vitro de viabilidade celular, observou-se que a morfologia celular não foi alterada, assim como a viabilidade e a capacidade de multiplicação das células. No ensaio in vitro de migração celular, quando comparada ao controle, a formulação com gel de babosa acelerou o processo de migração celular. Os resultados confirmaram a possibilidade de incorporar gel de babosa em hidrogéis de alginato obtendo formulações estáveis e adequadas ao uso tópico com potencial emprego como cicatrizante de feridas.pt_BR
