dc.description.abstractFew drugs are available against Chagas disease, caused by protozoa Trypanosoma cruzi. Most of these drugs cause severe side effects and there is no data on a drug with a 100% cure rate. Screening of new anti-T. cruzi compounds is initiated with in vitro tests against epimastigotes and intracellular amastigotes. Against amastigote forms, clinically relevant because are present in mammalian host, trypanocidal effect is mainly evaluated by microscopic count of residual parasites. Although reproducible, this technique is time-consuming and depends on experience of the microscopist. This work aims to develop a screening method for trypanocidal compounds through detection of fluorescence emitted by TcGFP. Epimastigotes of Y strain (108 parasites/ml) grown in LIT+10% FBS medium were transfected with plasmid pROCKGFPNeo and selected by serial dilution and G418 resistance antibiotic. After selection, epimastigotes showed homogeneous and stable GFP expression. GFP expression also remained stable in both trypomastigotes and intracellular amastigotes. Transfection did not affect the in vitro replication profile of epimastigotes, nor their susceptibility to benzonidazole. Furthermore, the strain was able to infect and replicate in Vero E6 cells. Through fluorimetry, there was a direct correlation between fluorescence intensity and concentration of epimastigote (R2=0.9986) and intracellular amastigotes (R2=0.9559) of TcGFP parasites. Aiming to validate the fluorimetric technique for trypanocidal compound screening, epimastigotes and cells containing intracellular TcGFP amastigotes were incubated with different concentrations of benzonidazole for 48 and 24 hours, respectively. The results showed that anti- epimastigotes tests were viable just to the compounds that cause lysis of the parasites, due to the fluorescence detecting of GFP retained in non-lysed unviable parasites. For intracellular amastigote forms, the proposed fluorimetric method can be applied, replacing the time-consuming microscopic count.pt_BR
dc.description.resumoAtualmente poucos fármacos estão disponíveis para o tratamento da doença de Chagas, causada pelo protozoário Trypanosoma cruzi. Destes fármacos, a maioria causa severos efeitos colaterais e nenhum deles apresenta 100% de cura parasitológica. A triagem de novos compostos anti-T. cruzi são iniciados com testes in vitro contra epimastigotas e amastigotas intracelulares. Para as formas amastigotas, clinicamente relevantes por apresentarem-se no hospedeiro mamífero, o efeito tripanocida é principalmente avaliado pela contagem microscópica de parasitos residuais. Apesar de reprodutível, esta técnica é morosa e depende da experiência do microscopista. Este trabalho objetiva a padronização de um método de triagem de compostos tripanocidas através da detecção de fluorescência emitida por TcGFP. Epimastigotas da cepa Y (108 parasitos/ml) cultivados em meio LIT+10%SFB foram transfectados com o plasmídeo pROCKGFPNeo e selecionados por diluição seriada e pelo antibiótico de resistência G418. Após seleção, os epimastigotas mostraram expressão de GFP homogênea e estável. A expressão da GFP também se manteve estável em tripomastigotas e amastigotas intracelulares. A transfecção não afetou os padrões de replicação in vitro dos epimastigotas, nem sua suscetibilidade ao benzonidazol e também não alterou a capacidade da cepa de infectar e de se replicar em células Vero E6. Através de fluorimetria foi verificado que há correlação direta entre intensidade de fluorescência e concentração de epimastigotas (R2=0,9986) e amastigotas intracelulares (R2=0,9559) de TcGFP. Para validar a técnica fluorimétrica para triagens de compostos tripanocidas, epimastigotas e células contendo amastigotas intracelulares de TcGFP foram incubados com diferentes concentrações de benzonidazol por 48 e 24 horas, respectivamente. Os resultados mostraram que para epimastigotas o método proposto restringe-se a teste de compostos que causem a lise dos parasitos, devido a possibilidade de detecção da fluorescência emitida pela GFP retida em parasitos inviáveis não lisados. Para as formas amastigotas intracelulares, o método fluorimétrico proposto pode ser aplicado, substituindo a morosa contagem microscópica.pt_BR
