dc.description.abstractRhizobia has different mechanisms for promoting plant growth, which are important for plant development and the sustainability of ecosystems. However, abiotic factors (i.g., pH, temperature, and the presence of xenobiotics) can affect symbiotic performance, according to the species/strain analyzed. Previous researches highlighting the occurrence of new species of rhizobia of Campos Gerais region in the Paraná state indicated that the legumes native to this ecosystem are nodulated by phylogenetically diverse species/strains of rhizobia. Therefore, this study aimed to characterize thirty- six rhizobia strains isolated from root nodules of native specimens of Mimosa spp. and Desmodium spp. concerning plant growth-promoting activities and responses to abiotic stresses. Strains growth was significantly affected by temperatures of 37°C and 40°C, concentrations of NaCl (1%, 2%, and 3%), and at pH 3.0. While, at pH 4.5 and 6.0 when compared to the control, we did not find significant differences. Such result may be related to the chemical soil conditions of origin of the host plants. Regarding tolerance to heavy metals, although neither of the strains had a positive response to Cu, all the same strains were able to grow at the evaluated concentrations of Zn. Concerning Al, even the highest concentrations tested (i.e., 2mM), 88% of the strains analyzed indicate resistance to these conditions. Neither strain reveal antagonistic activity against phytopathogens tested. Concerning phosphate solubilization activity, about 69.44% of the strains expressed positive results, with solubilization rates ranging from 0.13 to 1.22. Five of the strains produced different quantities of indolic compounds, of which four were isolated to D. barbatum. Our results indicate that abiotic factors significantly and differentially interfered in the growth of rhizobia, and emphasize the importance of physiological characterization in the selection of microorganisms.pt_BR
dc.description.resumoOs rizóbios possuem diferentes mecanismos de promoção do crescimento de plantas, importantes para o desenvolvimento vegetal e sustentabilidade dos ecossistemas. No entanto, fatores abióticos como pH, temperatura e presença de xenobióticos podem afetar o seu desempenho simbiótico de forma variável, de acordo com a espécie/estirpe. Levantamentos prévios apontam que as leguminosas nativas dos Campos Gerais do Paraná são noduladas por espécies/estirpes de rizóbios filogeneticamente variados, inclusive com a ocorrência de novas espécies. O objetivo deste estudo foi caracterizar trinta e seis estirpes de rizóbios isolados de nódulos radiculares de espécimes nativas de Mimosa spp. e Desmodium spp. quanto às suas atividades promotoras do crescimento de plantas e tolerância a estresses abióticos. O crescimento das estirpes foi significativamente afetado pelas temperaturas 37°C e 40°C, pelas concentrações de 1%, 2% e 3% NaCl e pelo pH 3,0; já em pH 4,5 e 6,0, não houve diferença significativa quando comparado com o controle. Pode-se relacionar tal resultado às condições químicas do solo de origem das plantas hospedeiras. Quanto à tolerância a metais pesados, nenhuma das estirpes apresentou resistência ao Cu nas três concentrações testadas enquanto todas foram capazes de crescer nas concentrações avaliadas de Zn. Quanto ao Al, 88% das estirpes apresentaram resistência à mais alta concentração testada (2mM). Nenhuma das estirpes apresentou atividade antagônica contra os fitopatógenos testados. Para a atividade de solubilização de fosfato 69,44% das estirpes tiveram resultados positivos, com índices de solubilização variando entre 0,13 a 1,22. Cinco das estirpes testadas produziram quantidades distintas de compostos indólicos, das quais quatro foram isoladas de D. barbatum. Nossos resultados indicam que fatores abióticos interferem de modo significativo e de forma diferencial no desenvolvimento dos rizóbios, demonstrando a importância da caracterização fisiológica na seleção de microrganismos.pt_BR
